Seeded Polymerization through the Interplay of Folding and Aggregation of an Amino-Acid-based Diamide

近年,超分子ポリマーの形成過程を時間で制御する精密超分子重合法の確立が進んでいる.このアプローチでは,超分子ポリマーを断片化した種(たね)や開始剤の添加により超分子重合を開始でき,ポリマー材料の根本的な構造要素である長さや長さ分布を制御することが可能である.長さの揃った超分子ポリマーは均質な物性の発現が見込まれ,機能性に優れると期待される.精密超分子重合法の実現には,モノマー分子の自発的な超分子ポリマー化が抑制された非平衡系を確立する分子設計が鍵となる.しかし,この重合法を適用できる化合物群には制限があり,汎用分子骨格の開拓が求められていた.今回我々は,分子内フォールディングと超分子ポリマー形成を促すアミノ酸ジアミド骨格に着目した.アミノ酸ジアミドに高発光性のピレンを導入し,種々のスペクトル測定と顕微鏡観察により超分子ポリマー形成過程を評価した.その結果,分子内水素結合によりジアミド骨格が折り畳まれた構造を形成し,超分子ポリマーの形成が一時的に抑制されることを見出した.また,得られた非平衡系を利用し,種の添加により超分子重合を開始させることに成功した.ジアミド骨格は自己組織化を促す汎用分子骨格であり,精密超分子重合の実現に向けた有用性を見出した点で重要な成果であると位置付けられる.
Amino-acid-based diamides are widely used as a substructure in supramolecular polymers, and also represent key components of polypeptides that help to understand protein folding. Herein, we report that the interplay of folding and aggregation of a diamide can be used to achieve seed-initiated supramolecular polymerization. For that purpose, a pyrene-substituted diamide was synthesized, in which pyrene is used as a tracer to monitor the supramolecular polymerization. Thermodynamic and time-dependent studies revealed that the folding of the diamide moiety, via the formation of intramolecular hydrogen bonds, effectively prevents a spontaneous nucleation that leads to supramolecular polymerization. Under such out-of-equilibrium conditions, the addition of seeds successfully initiates the supramolecular polymerization. These results demonstrate the utility of such amino-acid-based diamides in programmable supramolecular polymerizations.
